چکیده

A fluorometric-coupled reaction for the accurate and rapid determination malate, citrate, pyruvate and oxaloacetate is presented. The method was found useful for an accurate and rapid determination of these metabolites in low volumes of milk, yogurt, apple and lemon juice and wines without considerable pretreatment. In particular, this method was found valuable in characterising the outcome of maloactic acid fermentation (MLF) in wine and outlined for the first time fundamental differences in MLF between red and white wines. Thus, this method has merit in analysing these substances in heterogeneous, opaque and colorful foods. 2011 Elsevier Ltd. All rights reserved.

Application of immobilized citrate lyase in a bioanalytical system for determination of citrates

* corresponding author. e-mail: [email protected] A bioanalytical protocol based on the enzymatic-spectrophotometric micro-method for determination of citrate with immobilized citrate lyase (CL) is described. In the design of this spectrophotometric bioanalytical system, L-malate dehydrogenase (L-MDH), oxaloacetate decarboxylase (OACD) and L-lactate dehydrogenase (L-LDH) were applied as a biologica...

Blue native polyacrylamide gel electrophoresis and the monitoring of malate- and oxaloacetate-producing enzymes.

We demonstrate a facile blue native polyacrylamide gel electrophoresis (BN-PAGE) technique to detect two malate-generating enzymes, namely fumarase (FUM), malate synthase (MS) and four oxaloacetate-forming enzymes, namely pyruvate carboxylase (PC), phosphoenolpyruvate carboxykinase (PEPCK), citrate lyase (CL) and aspartate aminotransferase (AST). Malate dehydrogenase (MDH) was utilized as a cou...

Fluorometry of citrate in serum, with use of citrate (pro-3S)-lyase.

We describe a procedure for enzymatic assay of citrate in human serum. The citrate is degraded to acetate and oxaloacetate with citrate oxaloacetate-lyase (pro-3S-CH2-COO- yields acetate) (EC 4.1.3.6). Some oxaloacetate loses CO2 to form pyruvate. Addition of malate and lactate dehydrogenases (EC 1.1.1.37 and 1.1.1.27) permits determination of the oxaloacetate and pyruvate generated, and thus o...
